What Are Ion Selective Electrodes?
Ion Selective Electrodes (ISEs) are electrochemical sensors that produce a voltage proportional to the activity (effective concentration) of a specific ion in solution. They exploit membranes with selective ion permeability — the membrane potential follows the Nernst equation: E = E⁰ + (RT/nF) × ln(a_ion), where a_ion is the ion activity.
ISEs offer advantages over other analytical methods: no reagents required (after setup), direct in-sample measurement, field-portable designs, and fast response (seconds to minutes). They are essential for fluoride (no optical method is practical), and offer a practical alternative to ion chromatography for routine monitoring of nitrate, ammonium, chloride, and potassium.

Fluoride ISE — Critical Points
The lanthanum fluoride crystalline electrode is the only practical field method for direct fluoride measurement. Key operational requirements:
- TISAB (Total Ionic Strength Adjustment Buffer): Must be added to all standards and samples. TISAB serves three functions: (1) adjusts ionic strength to same level in all samples (linearizes Nernst response), (2) buffers pH to 5–5.5 (converts HF to F⁻, since ISE measures only free F⁻), (3) complexes interfering ions (Al³⁺, Fe³⁺, Si) with CDTA chelating agent. Without TISAB, results are meaningless.
- Interfering ions: OH⁻ (pH > 8 causes high reading), Al³⁺/Fe³⁺ (form AlF₃ complexes, reduce apparent fluoride) — both eliminated by TISAB addition
- Crystal maintenance: Rinse with deionized water between standards; never use abrasives
Nitrate ISE — Environmental Monitoring Applications
Nitrate ISEs with a liquid ion-exchange membrane are used for drinking water monitoring (WHO limit: 50 mg/L NO₃⁻), agricultural runoff screening, and aquaculture water quality. Key precautions:
- Interfering ions: Chloride (Cl⁻) is the primary interferent — ISA (ionic strength adjustor with ammonium sulfate) reduces chloride selectivity coefficient effect. At Cl⁻/NO₃⁻ ratios > 100, correction formulas must be applied.
- Reference electrode filling solution: Use double-junction reference to prevent Cl⁻ from reference junction contaminating the sample
- Membrane conditioning: Soak in 0.01 mol/L nitrate solution for 4 hours minimum before first use
Ammonium/Ammonia ISE — Wastewater and Aquaculture
Ammonium ISE (nonactin membrane) measures NH₄⁺; ammonia gas-sensing ISE measures total ammonia (NH₄⁺ + NH₃). Selection depends on application:
- Ammonium ISE: Better for wastewater below pH 8; measures ionic ammonium directly; interferences from K⁺ (K⁺/NH₄⁺ selectivity ratio ~0.1) must be accounted for in high-potassium samples
- Ammonia gas-sensing ISE: Sample adjusted to pH 11+ to convert all NH₄⁺ to NH₃(g) that diffuses through gas-permeable membrane; pH interference eliminated; better for low-concentration drinking water monitoring (0.01–0.1 mg/L)
ISE Calibration Methods
Two standard calibration approaches:
- Calibration curve method: 5-point standard curve (log concentration vs mV). Recommended for precise quantitative work. Check slope: should be 59.2 mV/decade for monovalent ions, 29.6 mV/decade for divalent ions (at 25°C). Slope < 50 mV/decade indicates degraded membrane.
- Standard addition method (SAM): Add known volume of concentrated standard to sample → measure mV shift → calculate ion concentration mathematically. Advantage: matrix matching is automatic. Preferred for complex matrices.
Heavy Metal ISEs (Lead, Copper, Cadmium)
Crystalline membrane ISEs for heavy metals use mixed sulfide pressed pellets (Ag₂S + PbS for Pb²⁺; Ag₂S + CuS for Cu²⁺). Primary applications: fast screening of smelter effluent, mining drainage, electroplating wastewater. Detection limits (without preconcentration): Pb²⁺ 0.02 mg/L, Cu²⁺ 0.01 mg/L, Cd²⁺ 0.01 mg/L. For regulatory compliance, confirm with AAS or ICP-MS.
