What Is TOC and Why Measure It?

Total Organic Carbon (TOC) is the amount of carbon bound in organic compounds dissolved or suspended in water, reported in mg/L or μg/L. TOC is a non-specific aggregate parameter — it measures the sum of all organic carbon, unlike COD or BOD which measure oxygen demand. TOC is preferred when: (1) the wastewater contains compounds that interfere with COD/BOD tests; (2) ultra-low detection limits are needed (ppb range); (3) real-time online monitoring is required.

Key industries using TOC: pharmaceutical (USP, EP water system validation), semiconductor (ultrapure water monitoring), power generation (boiler feedwater, condensate purity), environmental monitoring (surface water, groundwater, industrial effluent), food and beverage (CIP validation, product water testing).

TOC Measurement Methods

High-Temperature Combustion (HTC)

The sample is injected into a combustion tube at 680°C (with platinum catalyst) or 950–1200°C (high-temperature, no catalyst). All carbon is oxidized to CO₂, which is detected by non-dispersive infrared (NDIR) detector. HTC is the reference method (ISO 8245, Standard Methods 5310-B) with widest linear range (0.1 mg/L to 30,000 mg/L) and lowest detection limit achievable with proper optimization (2–5 μg/L with UV-persulfate pretreatment and ultra-low TOC standards).

UV/Persulfate Wet Oxidation

The sample is oxidized by UV irradiation plus persulfate (K₂S₂O₈) reagent at lower temperature (typically 100°C or ambient UV). Gentler oxidation — suitable for low-concentration samples (drinking water, pharmaceutical water). Less effective for refractory compounds (humic acids, certain industrial chemicals) and high-matrix samples. Lower maintenance than HTC combustion furnaces. Common in pharmaceutical and ultrapure water applications where concentrations are 0–2 mg/L.

  • Measurement range: Pharmaceutical/semiconductor: 0–500 μg/L (ppb range). Industrial wastewater: 0–1,000 mg/L. Select analyzer with auto-dilution for variable-concentration streams.
  • Oxidation method: UV/persulfate for ultra-pure water (cleaner, reagent-based); HTC combustion for complex industrial matrices (more robust oxidation).
  • Calibration requirements: Potassium hydrogen phthalate (KHP) is the primary TOC calibration standard (ISO traceable). Pharmaceutical online analyzers require 21 CFR Part 11-compliant audit trails and electronic records.
  • Response time: UV/persulfate online: 3–8 minutes per cycle. Combustion-based online: 5–15 minutes. For rapid process upsets, consider incorporating UV-Vis absorbance as a fast surrogate (response < 1 minute) with TOC correlation.

Pharmaceutical Water System TOC Requirements

USP <643> (Total Organic Carbon) sets the standard for pharmaceutical water TOC testing. Action level: 500 μg/L (0.5 mg/L) for purified water and water for injection (WFI). Online TOC monitoring with automated alarm is required in GMP-compliant water distribution systems. Key validation requirements: system suitability test using sucrose (easily oxidized reference standard) and 1,4-dioxane (refractory reference standard) — the online analyzer must recover both within acceptance criteria.

TOC vs COD Correlation

For consistent wastewater streams, TOC and COD are correlated: COD ≈ TOC × 2.67 for fully oxidized compounds (theoretical oxygen demand / carbon mass ratio). However, real correlation factors vary with wastewater composition (0.5 to 4.0 for different industrial streams). Establishing a site-specific TOC/COD correlation allows continuous TOC measurement to substitute for batch COD testing — reducing lab costs and providing real-time data. Minimum 20 paired TOC-COD measurements are recommended to establish a statistically valid correlation (R² > 0.95).
